Tyrosine synthesis in a Pseudomonas species.

Pseudomonas species ATCC 11299a synthesizes tyrosine via an inducible phenylalanine hydroxylase when grown on phenylalanine or tyrosine as a carbon source (G. Guroff and T. Ito, Biochim. Biophys. Acta 77:159, 1963; J. Biol. Chem. 240:1175, 1965). A second pathway to tyrosine synthesis involving transamination between glutamic acid or phenylalanine and p-hydroxyphenylpyruvic acid has been detected in this organism. p-Fluorophenylalanine, an inhibitor of phenylalanine hydroxylase, does not block the transaminase, and the latter activity may be demonstrated when there is reduced hydroxylase activity. The presence of this transaminase in glycerol-grown cells, in which no hydroxylase activity is demonstrable, indicates that it is a major pathway to tyrosine synthesis in this species. The growth medium has been described (G. Guroff and C. A. Rhodes, J. Biol. Chem. 242: 3644, 1967); carbon sources were either 0.2% phenylalanine or 1% glycerol. The high speed supernatant fraction (G. Guroff and T. Ito, and S. Udenfriend, J. Lab. Clin. Med. 50:733, 1957), and interference from pyridoxal phosphate, when present, was eliminated by using a zero-time sample as a control.
